
Mapping of Serologic Epitopes
Studies in this component of the workshop will be focused on defining HLA serologic epitopes contributing to antibody mediated rejection in renal and cardiac transplant recipients (because the pathology is best worked out in these groups).
Background:
The widespread use of highly sensitive and specific solid phase assays for HLA antibody definition has revealed that antibody reactivity is much more complicated than previously appreciated when only cytotoxic panels or ELISA techniques were used. Epitopes Pilot Project
The pilot project will evaluate testing performed by multiple laboratories using standardized SOPs of donor/recipient pairs where DSA is present. Samples, custom and commercial reagents will be provided by the Workshop organizers. Testing includes IgG SAB I, IgG SAB II, Bio-C1q SAB I, Bio-C1q SAB II.
Analysis of DSA Epitopes from Donor/Recipient Transplant Pairs
This project requires serum & DNA or complete HLA typing information by NGS HLA from donor/recipient pairs, first transplant, single organ (heart or kidney) where DSA is present but outcome data is not available. Solid phase testing to be performed using standardized work shop protocols, custom and commercial reagents including IgG SAB I, IgG SAB II, Bio-C1q SAB I, Bio-C1q SAB II.

Analysis of DSA Epitopes and FXM
This project requires serum, DNA & cells of donor/recipient pairs where DSA present, testing to be performed using standardized SOPs, custom and commercial reagents. Testing includes IgG SAB I, IgG SAB II, Bio-C1q SAB I, Bio-C1q SAB II and DSA-FXM, DNA or complete HLA typing information by NGS HLA from donor/recipient pairs. Analysis: DSA FXM, other FXM methods when available, pre and post-Transplant antibody results and transplant outcomes. Donor pairs may be from Kidney or Heart Transplant pairs but must be single organ primary Transplants.

Analysis of DSA Epitopes and Graft Outcomes
This project requires serum & DNA or complete HLA typing information by NGS HLA from donor/recipient pairs, first transplant, single organ (heart or kidney) where DSA present. Graft outcome information is required. Results obtained from various epitope analysis will be compared to solid phase results and NGS HLA typing from donor and recipient. Solid phase testing to be performed using standardized work shop protocols, custom and commercial reagents including IgG SAB I, IgG SAB II, Bio-C1q SAB I, Bio-C1q SAB II.
